The goal of the project is to develop an understanding of IL2 availability in CD8 T cell culture and its impact on culture performance in order to inform feeding strategies in scaled up processes for optimal culture performance.  We aim to accomplish this by building a mathematical model from empirical measurements of  IL2 degradation and consumption in culture and how modulating IL2 concentration impacts cell viability, diameter, growth rates, metabolism, and cell phenotype through two distinct phases of CD8 T cell growth.

  • Maintain extended cell cultures of CD8 T cells with sterile technique
  • Collect and analyze samples for IL2 concentration by ELISA
  • Learn and use appropriate lab equipment
  • Monitor and record VCD, viability, and cell diameter
