Title: R&D Biopharmaceuticals Summer Intern

Location: Gaithersburg, MD

Duration: 12 weeks

We are are seeking an undergraduate (ingoing junior or senior) or graduate student pursuing a degree in chemistry or biochemistry for a summer internship. 

At AstraZeneca, our ever-growing new modality biotherapeutics rebuild existing protein construct with innovative designs to fight disease in a more sophisticated way. These exciting new molecules yet impose greater challenges throughout the development process including product characterization. For instance, disulfide-bond related impurities, such as tri-sulfide linkages, disulfide-linked dimers, disulfide-scrambling have been observed. Cell-based assay reveals increased disulfide scrambling level could correlate with the decreased bioactivity. To better understand the impact of disulfide arrangement on the molecular function, advanced technologies such as hydrogen-deuterium exchange mass spectrometry (HDX-MS) and covalent labelling will be be applied to probe the conformational differences of the disulfide isoforms.

Primary Duties:

The intern will perform HDX-MS and covalent labelling experiment to pinpoint the regions that exhibit altered structural conformation. The analytical data will be incorporated with the protein 3D structure by computational modelling and the structural differences between the disulfide isomers can be visualized. Together with the bioactivity data from cell-based potency assays, the structural-function relationship of therapeutic protein molecules can be established.

 
Position Requirements
  • Hands-on lab experience (pipetting small volumes, creating dilutions) and principles of liquid chromatography and mass spectrometry.
  • An interst in biopharmaceuticalschemistry, analytical chemistry, and research.
  • Minimum GPA: 3.5
  • Currently enrolled fulltime in an academic program with the gool of seeking a minimum of a bachelor degree. 
  • Authorization to work in the US without sponsorship.
