What are the objectives/goals of the position?
Multi-attribute method (MAM) using a mass spectrometry (MS) has been adopted in our analytical BPD and showed promising benefits providing sensitive and selective information, and potential to reduce the number of arrays required for product quality control. Current peptide mapping method takes more than 6 hours of sample preparation time due to the necessary steps of in-solution tryptic digestion followed by buffer exchange, even the entire sample prep is automated using a robotic system.
A selected summer intern will evaluate several innovative approaches of protein digestion to reduce the prep time, in turn achieving faster turn-around-time of peptide mapping analysis, which could potentially be utilized in next generation manufacturing.
The goals of this project are following: 1. Evaluate and develop methods using various types of trypsin immobilized beads/magnetic beads from various venders; 2. Evaluate and optimize methods using the high throughput or automated devices; 3. Compare digestion efficiency and MAM focused mass spec data of peptide digests.
What are the primary duties of this position?
Working in the lab, evaluating sample preparation methods for peptide mapping liquid chromatography mass spectrometry analysis.
